I think this is quite a common thing on ae101's, mine did it as well. It's possible to get the wire back onto the pully, but not easily. Is the wire still connected to the triangular metal piece that actually moves up and down that the window is attached to? If it is it will be a lot easier to fix, though If it is attached at both ends (the window and the motor) it really shouldn't come off the lower pulley, because when tightened it doesn't get much room to move.
If it's still attached, like jayznz said, check its connection to the actual motor. I personally wouldn't recommend pulling off the circular metal (or in some cases plastic) casing cover on the spindle unless your absolutely sure nothing else could have caused it, as once off, if the wires dislodge, it is a real bitch to try and get them evenly arranged back into the grooves in the spindle, but like jayznz said, not impossible (though It's pretty $$% close!), so if not careful the net result is you need a new window unit.
But also like jayznz said sometimes something funky is going on inside there, so you may not have any choice. One cause is that the wires have begun to fray from the square metal stoppers. If this has happened, see how badly, and if it is possible to retwist the wire so it's width is still the same, because once it frays, it won't stay in the spindle grooves, thus stops the window and blah, once this happens, you generally need a new window motor.
